The density, genetic structure and diversity of airborne bacterial communities were assessed in the outdoor atmosphere. Two air samples were collected on the same location (north of France) at two dates (March 2003 (sample1) and May 2003 (sample 2)). Molecular culture-independent methods were used to characterise airborne bacterial communities regardless of the cell culturability. The automated-ribosomal intergenic spacer analysis (A-RISA) was performed to characterise the community structure in each sample. For both sampling dates, complex A-RISA patterns were observed suggesting a highly diverse community structure, comparable to those found in soil, water or sediment environments. Furthermore, differences in the genetic structure of airborne bacterial communities were observed between samples 1 and 2 suggesting an important variability in time. A clone library of 16S rDNA directly amplified from air DNA of sample 1 was constructed and sequenced to analyse the community composition and diversity. The Proteobacteria group had the greatest representation (60%), with bacteria belonging to the different subdivisions @a- (19%), @b-(21%), @c-(12%) and @d-(8%). Firmicute and Actinobacteria were also well represented with 14% and 12%, respectively. Most of the identified bacteria are known to be commonly associated with soil or plant environments suggesting that the atmosphere is mainly colonised transiently by microorganisms from local sources, depending on air fluxes.

Description:
Municipal solid waste (MSW) composts have been used to maintain the long-term productivity of agroecosystems and to protect the soil environment from overcropping, changes in climatic conditions and inadequate management; they also have the additional benefit of reducing waste disposal costs. Since MSW may contain heavy metals and other toxic compounds, amendments cannot only influence soil fertility, but may also affect the composition and activity of soil microorganisms. The effects of MSW compost and mineral N amendments in a 6-year field trial on some physical-chemical properties, enzyme activities and bacterial genetic diversity of cropped plots (Beta vulgaris-Triticum turgidum rotation) and uncropped plots were investigated. The compost was added at the recommended and twice the recommended dosage (12, 24tha^-^1). Amendments of cropped plots with MSW compost increased the contents of organic C from 13.3 to 15.0gkg^-^1 soil and total N from 1.55 to 1.65gkg^-^1 soil. There were significant increases in dehydrogenase (9.6%), @b-glucosidase (13.5%), urease (15.4%), nitrate reductase (21.4%) and phosphatase (9.7%) activities. A significant reduction in protease activity (from 3.6 to 2.8Ug^-^1 soil) was measured when a double dose of compost was added to the cropped plots. No dosage effect was detected for the other enzymes. Changes in the microbial community, as a consequence of MSW amendment, were minimal as determined using denaturing gradient gel electrophoresis, rDNA internal spacer analysis and amplified ribosomal DNA restriction analysis of bacteria, archaea, actinomycetes, and ammonia oxidizers. This indicates that there was no significant variation in the overall bacterial communities nor in selected taxonomic groups deemed to be essential for soil fertility.

A collection of 50 indigenous meta-toluate tolerating bacteria isolated from oil-contaminated rhizosphere of Galega orientalis on selective medium was characterized and identified by classical and molecular methods. 16S rDNA partial sequencing showed the presence of five major lineages of the Bacteria domain. Gram-positive Rhodococcus, Bacillus and Arthrobacter and gram-negative Pseudomonas were the most abundant genera. Only one-fifth of the strains that tolerated m-toluate also degraded m-toluate. The inoculum Pseudomonas putida PaW85 was not found in the rhizosphere samples. The ability to degrade m-toluate by the TOL plasmid was detected only in species of the genus Pseudomonas. However, a few Rhodococcus erythropolis strains were found which were able to degrade m-toluate. A new finding was that Pseudomonas migulae strains and a few P. oryzihabitans strains were able to grow on m-toluate and most likely contained the TOL plasmid. Because strain specific differences in degradation abilities were found for P. oryzihabitans, separation at the strain level was important. For strain specific separation (GTG)"5 fingerprinting was the best method. A combination of the single locus ribotyping and the whole genomic fingerprinting techniques with the selective partial sequencing formed a practical molecular toolbox for studying genetic diversity of culturable bacteria in oil-contaminated rhizosphere.

Brazil is the largest producer and consumer of the common bean (Phaseolus vulgaris L.), but yields are often low and may be improved by a higher N supply through symbiosis with rhizobia. One main limitation to the N"2-fixation process is the susceptibility of the symbiosis to environmental stresses frequent in the tropics, such as high soil temperatures and low soil moisture contents. Among other benefits, the no-tillage (NT) system reduces those stresses resulting in higher N"2 fixation rates and yields; however, the effects of NT on rhizobial diversity are poorly understood. This study evaluated the diversity of rhizobia compatible with common bean in cropping areas under the NT or the conventional tillage (CT) systems in Ponta Grossa, State of Parana, Southern Brazil. Genetic diversity was assessed by DNA analyses using the methodologies of BOX-PCR and RFLP-PCR of the 16S rDNA region. A high level of diversity was observed among the strains and the DNA profiles from the CT system were quite different from those from the NT system. Twenty-three RFLP-PCR profiles were obtained, indicating that many tropical rhizobial species remain to be described. Strain differentiation was achieved in the BOX-PCR analysis; diversity was slightly higher under the NT when compared with the CT system. Surprisingly, the rhizobial grouping based on cluster analysis of the RFLP-PCR of the 16S rDNA region indicated a higher diversity of species under the CT. It could be that the environmental stability offered by the NT system has led to a decrease in the number of species, with the predominance of the most successful ones, although genetic diversity within each species has increased. The results obtained in this study show that we still understand poorly the relation between microbial diversity and soil sustainability and that the complexity of the ecosystems require the evaluation of several parameters to define and monitor soil quality.

The phytotoxicity of tropospheric ozone at elevated concentrations can cause severe effects on plant vegetation. This study analyzed whether ozone-damaged plants transmit signals into the soil and thereby affect the structural diversity of bacteria inhabiting the rhizosphere. To generate highly stressful conditions, Malva sylvestris was raised from seeds under ozone exposure. Additionally, the plant age at the onset of ozone-stress was varied. Rhizosphere bacterial communities were analyzed by single-strand conformation polymorphism (SSCP) of PCR amplified partial 16S ribosomal RNA genes. None of these vigorous stress conditions caused a change in the genetic profiles targeting Bacteria with primers hybridizing to highly conserved regions in the 16S rRNA genes, despite visibly ozone-triggered injuries of the leaves and a reduced root biomass. Differences in the SSCP profiles however were observed between plants of different ages. In a more realistic scenario, plants raised in the greenhouse under normal atmospheric conditions were exposed to ozone episodes as they can occur during summer in industrialized countries of the northern hemisphere. To increase the sensitivity of detection, genetic profiles representing Alphaproteobacteria, Actinobacteria and Pseudomonas were analyzed separately in addition to those of Bacteria. None of the profiles showed any ozone-related alterations. In summary, the results of this study indicated that, even when the plants were visibly injured by ozone, the stress was not transmitted to the soil in a way that affected the structural diversity of the dominant rhizosphere bacterial community.

Peach tree replant disease, though reported on in the literature for more than two centuries, has yet to have its causes clearly defined. Decline in peach productivity has been attributed to toxic agents, insects, nutritional disturbances, spray residues, fungi and nematodes. Bacteria has also been indicated as a contributing factor. Peach replant disease was reproduced by using two successive cultures on the same soil. Bacterial communities were isolated and characterized from healthy and diseased peach trees. The potential role of cyanide production by rhizobacteria in the replant problem of peaches was studied. Culture-dependent (evaluation of the number of culturable bacteria, metabolic activities, Biolog^(R) GN2) and independent (ribosomal intergenic spacer analysis, RISA) methods were used, in order to compare bacterial community structure and diversity in healthy and sick soils and to evaluate the possible role of cyanide. Bacterial densities were significantly increased in sick soils. Metabolic activities (Biolog^(R) GN2) and genetic structure, observed through RISA, were also significantly modified in sick soils. Changes in the composition of individual microbial groups in the rhizosphere of peach trees excavated from healthy or sick soil indicated the involvement of rhizobacteria in the etiology of the replant sickness of peach soil. More than 60% of the strains isolated from healthy soils corresponded to Pseudomonas sp. and 58% of the isolates from sick soils were Bacillus sp. This study determined that Bacillus were able to produce in vitro HCN. It also appeared that in sick soil, there was a shift in the structure of bacterial communities with an increase noted in phytotoxic microorganisms capable of producing HCN compounds.

Biological nitrogen fixation plays a key role in agriculture sustainability, and assessment of rhizobial diversity contributes to worldwide knowledge of biodiversity of soil microorganisms, to the usefulness of rhizobial collections and to the establishment of long-term strategies aimed at increasing contributions of legume-fixed N to agriculture. Although in recent decades the use of molecular techniques has contributed greatly to enhancing knowledge of rhizobial diversity, concerns remain over simple issues such as the effects of sampling on estimates of diversity. In this study, rhizobia were isolated from nodules of plants grown under field conditions, in pots containing soil, or in Leonard jars receiving a 10^-^2 or a 10^-^4 serially-diluted soil inoculum, using one exotic (soybean, Glycine max) and one indigenous (common bean, Phaseolus vulgaris) legume species. The experiments were performed using an oxisol with a high population (10^5cellsg^-^1 soil) of both soybean rhizobia, composed of naturalized strains introduced in inoculants and of indigenous common-bean rhizobia. BOX-PCR was used to evaluate strain diversity, while RFLP-PCR of the ITS (internally transcribed spacer) region with five restriction enzymes aimed at discriminating rhizobial species. In both analyses the genetic diversity of common-bean rhizobia was greater than that of soybean. For the common bean, diversity was greatly enhanced at the 10^-^4 dilution, while for the soybean dilution decreased diversity. Qualitative differences were also observed, as the DNA profiles differed for each treatment in both host plants. Differences obtained can be attributed to dissimilarity in the history of the introduction of both the host plant and the rhizobia (exotic vs. indigenous), to host-plant specificity, rhizobial competitiveness, and population structure, including ease with which some types are released from microcolonies in soil. Therefore, sampling method should be considered both in the interpretation and comparison of the results obtained in different studies, and in the setting of the goals of any study, e.g. selection of competitive strains, or collection of a larger spectrum of rhizobia. Furthermore, effects of sampling should be investigated for each symbiosis.

excellent, easy to use guide.......2005-04-13
This is an excellent wildlife guide. Beautiful pictures, great maps, easy to follow directions to get to the viewing areas and easy to read text. This is currently my 6 year old son's favorite book (he reads above his grade level) and he is requesting other books in this series. He has learned a lot about the different animals that live in our home state and recognizes some of the places they list to view the animals.

it fell apart when i opened it, the stamps fell out no matter how many sticky things i put on behind, just, terrible!!! If you are reading this Mr. MacDonald, you should make some improvements. "A waste of Money," my kids say.

Nice Biography - Skips the minutiae and gets to the point.......2006-03-23
I bought this book because I thoroughly enjoyed the author's previous book "The Way You Wear Your Hat - Sinatra and the Lost Art of Livin'". This book was much shorter, even though it could be argued that its subject is deserving of perhaps even greater coverage. The most fascinating thing I learned while reading this book is that Sinatra and Hefner did not really bond...Frank was kinda upset that Hef had all the girls. The Mansion was the one place to which Sinatra could go and NOT find it to be all about Sinatra.
I knew very little about Hef until I read this book. I still don't know as much as I would have liked. This book is much more akin to what we would find out if Hef had written a dating profile on match.com (favorite movies, favorite food, favorite drink, etc). We learn very little about his formative years, other than that Hef's first great love did not love him back (but visits him at the mansion from time to time, even to this day). The author assumes that we know that Hef is wealthy, but we never get an idea of the degree of his wealth. I know that there is a magazine and a cable network....but younger readers may not know that there was also a television show, nightclubs, and a whole history extending back into the 1950's. These are only glanced over. It would have been nice to see more homage paid to the influence of Playboy on the shifting cultural attitudes during the 1960's.
I look forward to the author's upcoming Johnny Carson book and hope that it takes on the same flavor as these other two. Then the "holy trinity of cool" will be complete.
A little light on a fascinating man.......2005-10-10
Hef's Little Black Book isn't some story of Playboy the magazine or the empire, a biog of Hef or anything more than really a kind of puff-piece valentine from Hef to himself. I say from Hef because he's listed as the author, though "and Bill Zehme" presumably did most of the writing, organizing, interviewing, and editing.
However, given its parameters, the book works surprisingly well. Zehme did a similar book - a better one, though - on Sinatra a few years back, and its organization by subject/theme, its adoring fifties-style prose and please-pass-on-your-wisdom-o-master tone which strangely enough worked very well on the Sinatra piece is used again here. It's sort of effective. The book is a mixture of Zehme's narrative in the above voice, quotes from Hefner mixed in, and dozens of excellent photos of all types.
Hef passes on pearls of wisdom regarding women, romance, enjoying life and games, business, sex and the like. The Bed is covered in detail, with blueprints and everything. Much of it is not especially deep or new or earthshattering. I don't know that it really touches on what makes Hef such a fascinating figure or so important a man in 20th century life. But it's not uninteresting to Hefner aficionados.
What is in fact the goods on Hef is that he managed to first define the upscale male fantasy life, and then proceeded to insert himself into the picture and live it, for fifty nonstop years of uncompromising hedonism. In doing so he became a living symbol of the sexual revolution, and in the magazine's Playboy Philosophy he defended and explained his thinking brilliantly. It could not have worked without tremendous charm, business acumen (including the knowledge of when to step down from day-to-day operating control and let more capable managers take over), and self-control exercised over himself, and he surely kept very good people watching his back.
This book doesn't tell that story. If interested, there are many out there that do; I particularily like Russell Miller's Bunny, from back in the troublesome 80s. But this book does have fantastic and rarely seen photos from the 50s, 60s and 70s which make up for a lot, and one does get a faint glimpse of an unusual man.
Hef's little black book gives short and sassy glimpse of Hef........2005-09-09
You have to hand it to Hef. Well into his twilight years he manages to surround himself with beautiful, nubile young women who cater to his every whim.
In "Hef's Little Black Book" little glimpses of Hef's life are shared. We learn that Millie (his first wife) cheated on him, thus opening the door for his dalliances. We discover that the gorgeous Barbie Benton broke up with him when she discovered he was double dipping with another. That movie night is a big time in Hef's life. That black silk pj's make for good outerwear etc.
If you are a diehard Hef fan you may enjoy this book. On the other hand if you want to learn more about the playmates with shocking insights into Hef such as the awful reality that he does NOT use protection read Jill Ann Spaulding's book, "Upstairs".
Overall "Hef's Little Black Book" is indeed all about Hef, and only for those who want nitty gritty details shared by Mr. Hefner about himself.

Long ago I went to work for Playboy in Chicago and it was a thrill to be a part of Hefner's World. This book reminded me of why it was such fun. There is a mystique about this man and what he was able to do, living life on his own wide-eyed terms. I'm reminded here that he has always been the opposite of jaded--just a sunny funny smart revolutionary guy. It's nice to see it all spelled out, lest we forgot. The pictures took me back to a simpler happier time. What a trip.
