Description:
One of the popular ideas is that decline in methyl-directed mismatch repair (MMR) in carbon-starved bacteria might facilitate occurrence of stationary-phase mutations. We compared the frequency of accumulation of stationary-phase mutations in carbon-starved Pseudomonas putida wild-type and MMR-defective strains and found that knockout of MMR system increased significantly emergence of base substitutions in starving P. putida. At the same time, the appearance of 1-bp deletion mutations was less affected by MMR in this bacterium. The spectrum of base substitution mutations which occurred in starving populations of P. putida wild-type strain was distinct from mutation spectrum identified in MMR-defective strains. The spectrum of base substitutions differed also in this case when mutants emerged in starved populations of MutS or MutL-defective strains were comparatively analyzed. Based on our results we suppose that other mechanisms than malfunctioning of MMR system in resting cells might be considered to explain the accumulation of stationary-phase mutations in P. putida. To further characterize populations of P. putida starved on selective plates, we stained bacteria with LIVE/DEAD kit in situ on agar plates. We found that although the overall number of colony forming units (CFU) did not decline in long-term-starved populations, these populations were very heterogeneous on the plates and contained many dead cells. Our results imply that slow growth of subpopulation of cells at the expenses of dead cells on selective plates might be important for the generation of stationary-phase mutations in P. putida. Additionally, the different survival patterns of P. putida on the same selective plates hint that competitive interactions taking place under conditions of prolonged starvation of microbial populations on semi-solid surfaces might be more complicated than previously assumed.

I travel to San Francisco every year and I "never leave home without Betty & Pansy"! They get down to the nitty gritty and give you a feel of what each venue is like and the customers who frequent it. The "Review" is indispensible for any gay or lesbian going to the city by the bay.

How could I not LOVE IT!!! I will proudly tell you that I am the "Naomi" named in the book and that every story about every place mentioned is absolutely true!! I even know who the others are that are nicknamed in the various chapters...so watch it. Betty goes to great lengths to bring you the real flavor and spin in the City. Her reviews are brutally honest and they set a high mark for those wishing to make a good impression. This book is must for all those coming to the City. Good but dated........1998-07-25
This was an excellent and very comprehensive review of everything (and I mean everything) about the gay scene in San Francisco. Unfortunately, it was written in 1996 and is now quite out of date and needs revision.
The chapters on the bars and sex clubs are good as are the chapters on restaurants and places to stay but call ahead - many of the places mentioned have closed.
Still worth buying - just be a little cautious before getting yourself into a compromising position.

To Each Her Own.......2007-01-16
I thought, after reading one of the previous reviews, that I should go to the bookstore and look through the book before buying, but I needn't have worried. While there are a small handful of "jester"-type hats in this book (what hat book doesn't have an assortment of these?), mainly for children who usually enjoy the offbeat, I am overall pleased with the selection and variety. Several of the hats are quite elegant and sophisticated, with Nordic, Fair Isle, snowflake, and a wide variety of other designs made from a good assortment of yarns and colors to please every fashion palate. And being part of the Knitter's Dozen series, I feel comfortable that the instructions will be carefully "knitted through" and edited so I won't find myself stuck in the middle of a project.
The cover is deceiving.......2006-08-04
On my seemingly endless mission to get a good solid hat book, I thought my quest was over after seeing the cover of this one. How wrong I was. There is not one single hat in here that I would knit for anyone, not even an enemy. The layout is great, the readability is excellent, but whole book seems to be about how silly you can make the hat, rather than showing some good wearable ones. I was very disappointed, as you can probably tell.

I really like this book in content and presentation. Clear pictures! Best of all...stupid looking, court jester hats are kept to a minimum. Most of the patterns that are listed for children have adult instructions which I always value. I hate it when a lovely hat only has instructions for an 18 inch head.
The patterns cover a variety of yarn weights and techniques. Plenty of lovely sophisticated styles in this book to please almost any hat knitter.

It's probably safe to call this one a classic; it's just unfortunate that it's out of print and a bit hard to find.
Peter Adams comes at the subject of bonsai from the point of view of the trained artist, and his training shows through here. His emphasis on artistic method should be a welcome antidote to the oft-found notion that bonsai is little more than keeping alive a cute little tree in a pot.
In addition, this book has a wealth of hands-on information concerning soils, horticulture and strategies for long-term development of specimens. Keep in mind that few true bonsai are created in less than a few years, even when starting with specimens that already have a well-developed trunk and roots.
Adams has a special respect for, and treatment of, collected trees -- those taken from the wild.
This is a very good how-to book with lots of examples of how individual specimens were designed and developed.
This book is perhaps a little intimidating for the beginner; a novice might be better off starting with something like Herb Gustafson's "Bonsai Workshop," which gives a better basic overview of the art form but shares Adams's project-oriented approach. Or John Naka's "Bonsai Techniques" I & II, which have a wealth of very accessible information. Or even Peter Adams's "The Art of Flowering Bonsai," which has a lot of general information on developing trees as well as species-specific information related to certain blossoming kinds.
But if you find a used copy of "Bonsai Design" somewhere, buy it. If you don't read it now, you'll want to refer to it later.

This book has been the go-to treatise for raising children since 1990. Dr. Hausner's contemporary message applies to all families seeking assistance with the issues related to childrearing. A great read and very insightful.
A great book for parents at any income level!.......2005-02-20
IF YOU WANT TO IMPROVE YOUR CHILD'S MOTIVATION AND SELF-ESTEEM, THIS BOOK IS FOR YOU.
My husband and I, parents of three children (ages 7, 6, and 2) attended one of Dr. Hausner's speaking engagements in Coral Gables, Florida, and we were impressed by both her relaxed yet captivating speaking style as well as the content of what she had to say (definately (...) material). We were each given a copy of Children of Paradise, and I set to reading it right away. I am now on amazon to order 5 copies to pass along to my close parenting friends regardless of their income level. I plan to keep my highlighted, marked-up copy by my nightstand for easy reference for a long time. While the book contains some insightful strategies for wealthy families in handling inheritance and trusts - issues that are not present in most families in America - this book also provides some of the best parenting advice I have read regarding effective communication with your children, consistent discipline, and issues of control. In Children of Paradise, Dr. Hausner - without blame - discusses character traits of successful, "type A" personality parents and how these traits which have made them successful in their careers do not translate into successful parenting, but rather, can result in lowering a child's self esteem and motivation. I would argue that the same qualities can be found in parents of any income level coping with the competition and stress of daily life with children and careers during a time when we tend to demand perfection of ourselves and others. Dr. Hausner offers specific examples and techniques to improve how parents speak with and listen to their children - at any age - including teenagers. I have started to change the way I communicate with my children and I see results. In appropriate situations, I have stopped imposing strict punishments to control their behavior in favor of allowing my children to experience the natural consequences of their own failure to do what they need to do. (Not when it involves personal safety - obviously.) I am trying to stop telling them what to do most of the time (or just doing it for them) in the interest of expediency. I already see how my children are becoming more responsible with these changes and more confident. In short, this is a terrific book for any educated parent - career or stay at home, middle or upper class. I cannot recommend it highly enough.
Children of Paradise.......2000-04-01
In recent history with so much instant wealth created from techno-stocks, it gives great insights on how to handle wealth in your family. Wealth may solve some problems, but creates others for families and this book gives solutions to avoid the pitfalls. It is helpful in keeping children of wealth grounded. Lee Hausner has worked with many wealthy families in the Beverly Hills School District and has first-hand knowledge in dealing with the issues surrounding children and wealth.

Charming biography of America's premier wood engraver.......1997-03-31
This biography is written by family members of Cole,
who is regarded as America's premier white-line wood engraver. An autobiographical introduction starts this
colorful summary of Cole's life and work. From humble
immigrant beginnings, he rose to the top of his craft
by the end of the 19th century. The book is illustrated
with a selection of his finest work, largely from
his work on European masters. The text is of interest
to art historians, but also has a general appeal as an
intriguing biography of an accomplished engraver who
travelled Europe in search of masterpieces to render in wood. He then presented the essence of the paintings to the American audience through magazines such as Scribner's and Century.
